With the most efficient weapon ever designed being only 5.2 megatons per tonne of material (B41 nuclear bomb - Wikipedia) we can actually get a close estimate of how massive that missile has to be.
Since we want 10,000 megatons we can multiply that number by 5.2 and get... 52 thousand metric tons of uranium/plutonium alone.
