To expand on the question

1) The game map is currently 25 system player starting clusters and a 100 system shared core cluster where players will eventually encounter each other. Is this large enough or does it need to be bigger?

I should have expected that really
I'd say its a good start. There can always be the option to expand in the future, and I think that may be wise, but I'd probably give it a while at this scale to test the rules and mechanisms first. Expand once the basics are shown to be fine.

If we have 25*10 = 250 systems worth of starting clusters, then a 100 system core sounds much too tiny -- if the "PvP area" is so much smaller than everyone's back yards, why is it even worth fighting over?

I would suggest that the core needs to be a multiple of the starting clusters, even if this means the starting clusters need to be shrunk a little in order to keep the core of manageable size.
